Biological Background: IFT88 Function and Localization

• IFT88 is a core component of intraflagellar transport (IFT) complexes and positively regulates primary cilium biogenesis. Related references: PMID:17604723
• Involved in autophagy, as it is required for trafficking of ATG16L and the expansion of the autophagic compartment.
• Localizes to centrosome, centriole, cilium basal body, ciliary axoneme, cytoplasm, and flagellum.
• Contains tetratricopeptide repeat (TPR) motifs that mediate protein-protein interactions essential for IFT complex assembly.
• Expressed in heart, brain, liver, lung, kidney, skeletal muscle, and pancreas.
• Alternative splicing generates multiple isoforms (based on the Alternative splicing keyword).

Experimental Guidance and Technical Tips

• The immunogen corresponds to amino acids 534–833 near the C-terminus; consider that this region may be present in most isoforms but verify in your system.
• For Western blot (WB), a major band near the predicted molecular weight of ~93 kDa is expected. Additional bands may appear due to post-translational modifications or degradation; validate with positive and negative controls.
• For immunofluorescence/immunocytochemistry (IF/ICC), optimize fixation and permeabilization to access ciliary and centrosomal compartments.
• In ELISA, the antibody can be used as a capture or detection reagent; check cross-reactivity with samples from other species beyond human, mouse, and rat if needed.
