Biological Background: IFNL3 Function and Localization
- IFNL3 (interferon lambda-3), also known as IL-28B/IL-28C, is a cytokine belonging to the type III interferon family.
- It plays a critical role in antiviral host defense, predominantly in epithelial tissues, by binding to the heterodimeric receptor IL10RB/IFNLR1 and activating the JAK/STAT signaling pathway to induce interferon-stimulated genes (ISGs).
- The receptor IFNLR1 has epithelial cell-specific expression, resulting in restricted targets and cell-type selective action of IFNL3.
- IFNL3 is involved in Toll-like receptor (TLR)-induced antiviral defense and contributes to the antiviral immune response in the intestinal epithelium.
- It also exerts immunomodulatory effects, including up-regulation of MHC class I antigen expression.
- The protein is secreted and contains a signal peptide and disulfide bonds, with a molecular weight of approximately 21.7 kDa.
- Alternative names include cytokine Zcyto22, interleukin-28B, and interleukin-28C.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminal region (aa 1-100) of human IFNL3. This peptide sequence may influence antibody specificity; validate in the relevant sample system.
- For Western blot, an expected band at ~22 kDa can be observed; consider using human or mouse epithelial cell lysates.
- Given the polyclonal nature and cross-reactivity with mouse, careful titration and inclusion of appropriate controls (e.g., peptide blocking) are recommended to confirm specificity.
- For ELISA, the antibody may be used as a capture or detection reagent; pairing with a monoclonal antibody recognizing a distinct epitope could enhance assay performance.
- Always optimize concentrations and conditions in your specific experimental setup.
