Biological Background: IFNGR1 Function and Localization
- Interferon gamma receptor 1 (IFNGR1) is the ligand-binding alpha chain of the interferon gamma receptor, also known as CD119.
- It plays a critical role in antimicrobial, antiviral, and antitumor immune responses by activating effector immune cells and enhancing antigen presentation (PubMed:20015550). Related references: PMID:20015550

- Upon interferon gamma binding, the intracellular domain of IFNGR1 undergoes a conformational change, enabling the recruitment and activation of JAK1 and JAK2 kinases, which phosphorylate IFNGR1 to create a docking site for STAT1 (PubMed:28883123). Related references: PMID:28883123
- The protein is localized on the cell membrane and features immunoglobulin-like domains, disulfide bonds, and glycosylation sites. It undergoes alternative splicing and is subject to phosphorylation and ubiquitin conjugation.
- Mutations in IFNGR1 are associated with primary immunodeficiencies, such as Mendelian susceptibility to mycobacterial disease (MSMD) (key word: Disease variant).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the intracellular domain (aa 270-489) of human IFNGR1; consider that the antibody may recognize both full-length and any truncated isoforms arising from alternative splicing.
- For Western blot, the predicted molecular weight is approximately 54 kDa, but glycosylation may result in a slightly higher apparent molecular weight.
- For immunohistochemistry on paraffin-embedded sections, optimization of antigen retrieval may be necessary.
- As a polyclonal antibody, lot-to-lot variability may occur; include appropriate positive and negative controls in each experiment.
- Cross-reactivity with mouse and rat IFNGR1 has been reported; validate specificity in your model system.
