Biological Background: IFNGR1 Function and Localization
- Interferon gamma receptor 1 (IFNGR1), also known as CD119 or IFN-gamma-R-alpha, is a single-pass type I membrane protein and a critical subunit of the interferon gamma receptor complex.
- Serves as the ligand-binding receptor for interferon gamma (IFNG), playing a key role in antimicrobial, antiviral, and antitumor immune responses by activating effector cells and enhancing antigen presentation. Related references: PMID:20015550
- Associates with the accessory factor IFNGR2 to form a functional receptor complex upon ligand binding. Related references: PMID:10986460, PMID:2971451, PMID:7615558
- Upon IFNGR1 engagement, the intracellular domain undergoes conformational changes allowing association of JAK1 and JAK2 kinases; activated JAK1 phosphorylates IFNGR1 to create a docking site for STAT1, leading to STAT1 phosphorylation, dimerization, nuclear translocation, and target gene transcription. Related references: PMID:28883123
- STAT3 can also be activated through IFNGR1, though the response is typically weaker.
- The intracellular domain phosphorylation also provides a docking site for SOCS1, which negatively regulates JAK-STAT signaling by competing with STAT1 binding (By similarity).
- Expressed on the cell membrane.
- Keywords: Glycoprotein, disulfide bond, alternative splicing, disease variant, phosphoprotein, and Ubl conjugation, highlighting its regulatory complexity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the intracellular region (aa 270–489), suggesting the antibody may detect total IFNGR1 protein by Western blot regardless of ligand binding or membrane localization.
- For Western blot, a band around 54 kDa is expected; consider using reducing conditions and appropriate controls (e.g., IFNGR1‑knockout validated cell lysates) to confirm specificity.
- The antibody is recommended for ELISA; ensure appropriate coating buffers and detection systems are optimized for the target protein.
- Cross‑reactivity with mouse and rat IFNGR1 has been reported; verify performance in the relevant species and tissue types before quantitative analyses.
