Biological Background: IFNAR1 Function and Localization
-
IFNAR1 (Interferon alpha/beta receptor 1), also known as Cytokine receptor class-II member 1 (CRF2-1), forms a heterodimeric receptor with IFNAR2 for type I interferons (IFN-α, IFN-β, IFN-ε, IFN-ω, and IFN-κ). Related references: PMID:10049744, PMID:14532120, PMID:15337770
-
Upon interferon binding, IFNAR1 and IFNAR2 proximity triggers cross-phosphorylation of associated Janus kinases (TYK2 on IFNAR1, JAK1 on IFNAR2), phosphorylation of receptor tyrosine residues, recruitment and phosphorylation of STAT transcription factors, and their nuclear translocation to regulate interferon-stimulated gene expression. Related references: PMID:21854986, PMID:32972995, PMID:7665574, PMID:7526154, PMID:19561067
-
IFNAR1 can also act independently of IFNAR2 as a functional receptor for IFNB1, activating a non-JAK-STAT signaling pathway.
-
Subcellular localization includes the cell membrane, late endosomes, and lysosomes, consistent with receptor internalization and trafficking.
-
IFN receptors are ubiquitously expressed. Multiple isoforms exist: isoform 1, 2, and 3 are detected in IFN-sensitive myeloma U266B1 cells, while only isoforms 2 and 3 are found in IFN-resistant U266R cells.
-
IFNAR1 is a member of the class II cytokine receptor family, possessing extracellular fibronectin type III repeats and a single transmembrane helix.
-
Key post-translational modifications include glycosylation, palmitoylation, phosphorylation, and ubiquitination, along with disulfide bond and isopeptide bond formation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal intracellular domain (aa 470–557), making the antibody suitable for applications like Western blotting (WB) and immunohistochemistry (IHC) that detect total IFNAR1 regardless of extracellular domain shedding or glycosylation state.
- For immunofluorescence (IF/ICC), permeabilization is necessary to allow antibody access to the intracellular epitope; include appropriate negative controls to verify specificity.
- As a polyclonal antibody, it may recognize multiple epitopes, enhancing sensitivity but potentially increasing background; consider optimizing blocking and washing conditions.
- Confirmed cross-reactivity with mouse and rat IFNAR1, but sequence homology in the immunogen region should be verified; validation in the relevant model system is recommended.
- For ELISA applications, ensure that the coating antigen matches the immunogen region or full-length protein; a sandwich ELISA format may improve signal-to-noise ratio.
- Keep cell/tissue lysates and reagents on ice during preparation to minimize proteolytic degradation of the ~64 kDa target.
