Biological Background: IFN gamma Function and Localization
- Interferon gamma (IFN gamma) is a type II interferon, also known as immune interferon, encoded by the IFNG gene in humans. It is secreted primarily by activated T lymphocytes and natural killer cells.
- Functions as a potent cytokine in antimicrobial, antiviral, and antitumor responses by activating effector immune cells and enhancing antigen presentation. Related references: PMID:16914093 PMID:8666937
- Signals through the JAK-STAT pathway upon binding to its receptor IFNGR1, leading to STAT1 activation and transcription of IFN gamma-regulated genes such as IRF1. Related references: PMID:8349687 PMID:16914093
- Enhances MHC class I antigen presentation by inducing immunoproteasome subunits and the activator PA28, increasing the quantity and quality of peptides for MHC loading. Related references: PMID:8666937 PMID:8163024 PMID:11112687
- Up-regulates MHC class II complex expression by promoting key molecules such as cathepsins B, H, and L. Related references: PMID:7729559
- Involved in the regulation of hematopoietic stem cells, affecting their development, quiescence, and differentiation during homeostasis (By similarity).
- Subcellular localization is secreted, consistent with its role as a cytokine. The pro-protein contains a signal peptide and undergoes cleavage on a pair of basic residues to generate the mature form.
- Post-translational modifications include glycosylation and pyroglutamic acid formation, as indicated by UniProt keywords.
Experimental Guidance and Technical Tips
- The immunogen spans the mature IFN gamma sequence (aa 24-166), targeting the functional domain; consider this when designing blocking peptides or competition assays.
- For Western blot, the predicted molecular weight is 19 kDa; ensure appropriate gel percentage and transfer conditions for low molecular weight proteins.
- Since IFN gamma is a secreted cytokine, sample preparation should consider its presence in serum, plasma, or cell culture supernatants; include appropriate controls and protease inhibitors to prevent degradation.
- For ELISA, this polyclonal antibody can be used as either capture or detection antibody; however, pairing with a monoclonal antibody for sandwich ELISA may improve specificity.
- Validate performance in relevant biological samples (e.g., activated T-cell lysates or stimulated PBMCs) to confirm reactivity and to establish optimal working dilutions empirically.
