Biological Background: IFN gamma Function and Localization
- Interferon gamma (IFNγ) is a type II interferon produced by immune cells like T-cells and NK cells, playing crucial roles in antimicrobial, antiviral, and antitumor responses by activating effector cells and enhancing antigen presentation. Related references: PMID:16914093, PMID:8666937
- IFNγ signals primarily through the JAK-STAT pathway after binding to its receptor IFNGR1, leading to STAT1 activation and transcription of IFNγ-regulated genes, including many transcription factors like IRF1. Related references: PMID:8349687, PMID:16914093
- It enhances class I MHC antigen presentation by inducing a switch to immunoproteasome catalytic subunits and up-regulating the activator PA28, improving peptide quantity, quality, and repertoire for MHC I loading. Related references: PMID:8666937, PMID:8163024, PMID:11112687
- IFNγ also up-regulates MHC class II surface expression by promoting the expression of cathepsins B, H, and L. Related references: PMID:7729559
- This secreted cytokine is predominantly released from activated T lymphocytes and influences hematopoietic stem cell development, quiescence, and differentiation.
- The canonical full-length IFNγ protein (NP_000610.2) has a predicted molecular weight of ~19 kDa; it undergoes proteolytic cleavage between basic residues, contains a signal peptide, is N-glycosylated, and features a pyrrolidone carboxylic acid modification.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 24–161 of human IFNγ (NP_000610.2), representing the mature secreted protein without the signal peptide. This design may provide specificity for the functional form of IFNγ in immunoassays.
- For Western blot (WB), consider the predicted molecular weight of ~19 kDa and potential shifts due to glycosylation. Use appropriate reducing conditions and validate band specificity in your sample system.
- For ELISA, this monoclonal antibody could be used as capture or detection reagent; pairing with a validated antibody against a non-overlapping epitope is recommended. Titration and matrix compatibility experiments are advised.
- This antibody is listed as cross-reactive with human IFNγ; cross-reactivity with other species is not guaranteed and should be independently verified if non-human samples are tested.
- Product shipped on ice; store at −20°C upon receipt to maintain stability. Avoid repeated freeze-thaw cycles.
