Biological Background: IFN gamma Function and Localization
- Interferon gamma (IFN-γ), also known as Immune interferon, is a type II interferon cytokine with pivotal roles in innate and adaptive immunity.
- It is primarily secreted by activated T lymphocytes and natural killer (NK) cells, exerting broad antimicrobial, antiviral, and antitumor activities.
- IFN-γ signals through the JAK-STAT pathway upon binding to its receptor IFNGR1, leading to STAT1 activation and transcription of downstream effector genes. Related references: PMID:16914093 PMID:8349687
- A key function is enhancement of antigen presentation: it induces immunoproteasome subunits and the PA28 activator to improve peptide generation for MHC class I loading, and upregulates MHC class II molecules via cathepsins. Related references: PMID:8666937 PMID:8163024 PMID:11112687 PMID:7729559
- The protein is secreted and undergoes post-translational modifications including glycosylation and signal peptide cleavage, yielding a mature molecular weight of approximately 19 kDa.
- IFN-γ also participates in hematopoietic stem cell regulation, influencing development, quiescence, and differentiation (By similarity).
Experimental Guidance and Technical Tips
- The immunogen encompasses amino acids 24–161 of human IFN gamma (NP_997468.1), covering the mature secreted protein; the antibody may recognize both precursor and processed forms.
- For Western blot (WB), the predicted weight is 19 kDa, but glycosylation can shift the apparent band higher; validate with appropriate controls.
- In IHC-P, optimize antigen retrieval and antibody dilution; reported cross-reactivity with mouse and rat should be confirmed in the relevant tissue context.
- For ELISA, this polyclonal antibody can be used as a capture or detection reagent; pairing with a validated monoclonal antibody is recommended for enhanced sensitivity.
- As a polyclonal preparation, lot-to-lot variability is possible; perform pilot testing for each new lot.
