Biological Background: IFN-gamma Function and Localization
- IFN-gamma (Interferon gamma, also known as Immune interferon) is a type II interferon encoded by the IFNG gene in humans.
- It is primarily secreted by activated T lymphocytes and NK cells and plays a pivotal role in antimicrobial, antiviral, and antitumor immunity by activating effector immune cells and enhancing antigen presentation. Related references: PMID:16914093 PMID:8666937
- Upon binding to its receptor IFNGR1, IFN-gamma triggers the JAK-STAT signaling cascade, leading to STAT1 activation, nuclear translocation, and transcription of interferon-regulated genes, including transcription factors like IRF1. Related references: PMID:8349687 PMID:16914093
- It enhances antigen presentation by inducing immunoproteasome subunits and the PA28 activator, thereby increasing the quantity and quality of peptides for MHC class I loading. Related references: PMID:8666937 PMID:8163024 PMID:11112687
- IFN-gamma also upregulates MHC class II expression by promoting cathepsin B, H, and L expression, further facilitating immune recognition. Related references: PMID:7729559
- Additionally, it participates in hematopoietic stem cell regulation, affecting development, quiescence, and differentiation.
- Keywords include antiviral defense, cytokine, secreted, signal, and glycoprotein.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the mature form of human IFN-gamma (amino acids 24–166), suggesting the antibody likely recognizes both native and denatured conformations, making it suitable for western blotting under reducing conditions.
- For flow cytometry intracellular staining, proper cell fixation and permeabilization protocols must be optimized, as IFN-gamma is a secreted protein and may require stimulation and secretion inhibitors for reliable detection.
- In ELISA applications, the antibody may perform as either capture or detection reagent; titration and pairing experiments are recommended to establish optimal working concentrations.
- Cross-reactivity with other species has not been reported; validation in non-human systems should be conducted if necessary.
