Biological Background: IFN-beta Function and Localization
- IFN-beta (also known as fibroblast interferon, IFB, IFF, IFNB) is encoded by the IFNB1 gene in humans.
- It is a type I interferon cytokine that plays a key role in the innate immune response to infections, developing tumors, and other inflammatory stimuli. Related references: PMID:10049744, PMID:10556041, PMID:6157094
- Signaling occurs through binding to the heterodimeric receptor IFNAR1/IFNAR2, activating the canonical JAK-STAT pathway to regulate transcription of interferon-regulated genes. Related references: PMID:10049744, PMID:10556041, PMID:7665574
- Beyond antiviral and antibacterial activities, IFN-beta regulates B-cell development, myelopoiesis, and LPS-inducible production of tumor necrosis factor.
- In the nervous system, IFN-beta promotes neuronal autophagy and clearance of alpha-synuclein, helping protect dopaminergic neurons.
- Compared to interferon-alpha, IFN-beta is more potent in inducing apoptotic and antiproliferative pathways required for tumor cell growth control.
- IFN-beta is a secreted glycoprotein containing disulfide bonds and phosphorylation sites, as indicated by the keywords: Secreted, Glycoprotein, Disulfide bond, Phosphoprotein.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 22-187 of mature human IFN-beta, so the antibody is expected to detect the secreted, mature form. Consider using reducing SDS-PAGE conditions for WB.
- For WB, validate the antibody in appropriate human cell lines or stimulated samples known to express IFN-beta (e.g., virus-infected fibroblasts).
- In ELISA, consider pairing with another anti-IFN-beta antibody or using recombinant IFN-beta as standard for quantitative detection.
- Optimal working dilutions for each application should be determined empirically by the end user.
