Biological Background: IFITM3 Function and Localization
- IFITM3 (Interferon-induced transmembrane protein 3) is a member of the interferon-inducible transmembrane (IFITM) protein family, also known as dispanin subfamily A member 2b (DSPA2b) or 1-8U.
- It functions as an IFN-induced antiviral protein that disrupts intracellular cholesterol homeostasis, inhibiting viral entry by preventing fusion with cholesterol-depleted endosomes. Related references: PMID:26354436 PMID:33239446 PMID:33270927
- Active against a broad range of viruses including influenza A, SARS coronaviruses (SARS-CoV and SARS-CoV-2), Ebola, Marburg, Dengue, West Nile, HIV-1, HCV, and VSV. Related references: PMID:26354436 PMID:33239446 PMID:33270927
- Plays a critical role in vacuolar ATPase (v-ATPase) structural stability and function, establishing endosomal pH necessary for the antiviral state. Related references: PMID:26354436
- Subcellular localization includes cell membrane, late endosome membrane, early endosome membrane, lysosome membrane, and perinuclear cytoplasm.
- Post-translational modifications include palmitoylation, phosphorylation, and ubiquitination, which may regulate its antiviral activity.
- In hepatocytes, coordinates with other IFITM proteins to restrict HCV infection by targeting virions for lysosomal degradation. Related references: PMID:26354436
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal amino acids 1–57 of human IFITM3, suggesting the antibody may recognize an epitope in this region.
- For Western blot (WB), the predicted molecular weight is approximately 15 kDa; however, post-translational modifications may cause shifts, so validate in the relevant sample system.
- IFITM3 expression is interferon-inducible; consider using interferon-treated human cell lines or tissues to enhance detection sensitivity.
- For IHC-P, optimize antigen retrieval and antibody dilution using appropriate controls, as detection may depend on expression levels and tissue processing.
- While validated for human reactivity, cross-reactivity with other species or IFITM family members is possible; verify in the intended experimental system.
