Biological Background: IFIT3 Function and Localization

  • IFIT3, also known as interferon-induced protein with tetratricopeptide repeats 3, CIG49, ISG-60, IFI-60K, IFIT-4, RIG-G, or P60, is an interferon-stimulated gene (ISG) member of the IFIT family.
  • Unlike some other IFITs that directly bind viral RNA, IFIT3 primarily acts as an adaptor protein, bridging TBK1 to MAVS on mitochondria, which leads to TBK1 activation and subsequent IRF3 phosphorylation to drive antiviral gene expression.
  • IFIT3 also exhibits antiproliferative effects by upregulating the cell cycle inhibitors p21 and p27; it sequesters COPS5 in the cytoplasm, preventing COPS5-mediated nuclear export and degradation of p27, while also suppressing MYC to relieve repression of p21.
  • IFIT3 can negatively regulate the apoptotic effects mediated by IFIT2, suggesting a modulatory role in programmed cell death.
  • Subcellular localization is primarily cytoplasmic, but IFIT3 can translocate to mitochondria where it interacts with MAVS.
  • IFIT3 protein is highly expressed in immune tissues such as spleen, lung, leukocytes, lymph nodes, placenta, bone marrow, and fetal liver; its expression is markedly elevated in PBMCs and monocytes from systemic lupus erythematosus (SLE) patients compared to healthy controls.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to an internal region (amino acids 128–331) of mouse IFIT3; the antibody is expected to recognize full-length IFIT3 protein, but epitope exposure may vary across assay formats.
  • For Western blotting, a band at ~56 kDa is predicted; verify specificity using interferon-treated and untreated cell lysates from human, mouse, or rat origin.
  • In immunohistochemistry (IHC-P), optimize antigen retrieval and dilution; cytoplasmic and mitochondrial staining should be anticipated based on known localization.
  • For ELISA, use as capture or detection antibody after pairing validation with appropriate standards; cross-reactivity with other IFIT family members has not been tested and should be assessed if needed.
