Biological Background: IFIT3 Function and Localization

  • IFIT3 (Interferon-induced protein with tetratricopeptide repeats 3), also known as CIG49, ISG-60, IFI-60K, IFIT-4, or RIG-G, is a 56 kDa cytoplasmic and mitochondrial protein induced by interferon.
  • It acts as an antiviral protein by enhancing MAVS-mediated signaling: IFIT3 bridges TBK1 to MAVS, promoting TBK1 activation and IRF3 phosphorylation, which then translocates to the nucleus to induce antiviral gene transcription.
  • It exhibits antiproliferative activity by up-regulating cell cycle inhibitors CDKN1A/p21 and CDKN1B/p27, partly by sequestering COPS5 in the cytoplasm to prevent p27 degradation, and by down-regulating MYC, a repressor of p21.
  • IFIT3 can negatively regulate the apoptotic effects of IFIT2.
  • Expression is elevated in peripheral blood mononuclear cells and monocytes from systemic lupus erythematosus patients, and it is also expressed in spleen, lung, leukocytes, lymph nodes, placenta, bone marrow, and fetal liver.
  • It belongs to the tetratricopeptide repeat (TPR) family and is involved in innate immunity and antiviral defense; the protein is subject to phosphorylation (at protein level) and has been identified in proteomics studies.

  • For western blot (WB), a major band around 56 kDa is expected. Consider using reducing conditions and appropriate positive control lysates (e.g., IFN‑treated HeLa or RAW 264.7 cells) to confirm specificity.
  • In ELISA, titrate the antibody to determine the optimal working concentration for your specific plate format and antigen. Validate with a standard curve using recombinant IFIT3 protein if available.
