Biological Background: IFI35 Function and Localization
- IFI35 (Interferon-induced 35 kDa protein, also known as IFP35) is encoded by the IFI35 gene; alternative splicing produces multiple isoforms.
- In response to interferon-alpha, IFI35 forms a complex with NMI, preventing proteasome-mediated degradation and correlating with dephosphorylation. This complex inhibits virus-triggered type I interferon/IFN-beta production and negatively regulates NF-kappa-B signaling by blocking nuclear translocation of p65/RELA. - Extracellularly, macrophage-secreted IFI35 acts as a damage-associated molecular pattern (DAMP), activating NF-kappa-B in adjacent macrophages via Toll-like receptor 4 (TLR4) to promote pro-inflammatory cytokine release. Related references: PMID:29038465
- Subcellularly, IFI35 localizes to the cytoplasm, nucleus, and can be secreted.
- It is expressed in a wide range of cell types, including fibroblasts, macrophages, and epithelial cells.
