Biological Background: IFI30 Function and Localization

• IFI30 (GILT, IP30) is a gamma-interferon-inducible lysosomal thiol reductase that catalyzes the reduction of protein disulfide bonds, playing a critical role in antigen processing and presentation for both MHC class I and class II pathways. Related references: PMID:10639150
• The enzyme contains a redox-active CXXC motif; the N-terminal cysteine initiates nucleophilic attack on substrate disulfide bonds, forming a mixed disulfide intermediate, which is resolved by the second cysteine to release reduced substrate and oxidized enzyme.
