Biological Background: IFI27 Function and Localization
- IFI27, also known as ISG12A, is an interferon alpha-inducible protein encoded by the IFI27 gene. Alternative splicing produces multiple isoforms.
- Functions as an adapter protein in type-I interferon-induced and TNFSF10-induced apoptosis. Promotes cytochrome C release from mitochondria, activating BAX and caspases 2, 3, 6, 8, and 9. Related references: PMID:18330707 PMID:24970806 PMID:27673746
- Plays a role in innate antiviral immunity against hepatitis C virus (HCV) by promoting ubiquitin-mediated proteasomal degradation of the viral NS5A protein via SKP2, and induces virus-induced pyroptosis by activating CASP3 after TRIM21-mediated 'Lys-6'-linked ubiquitination. - The immunogen corresponds to the N-terminal region (aa 1-100) of human IFI27. This region exhibits high sequence conservation across species, and cross-reactivity with rat has been demonstrated. For other species, perform in silico alignment to predict reactivity.
- For Western blot, a specific band at approximately 12 kDa is expected in IFI27-expressing samples. Include appropriate positive and negative controls to confirm specificity.
- In ELISA, the antibody can serve as either a capture or detection reagent. Optimize coating and detection conditions empirically.
- As a polyclonal antibody, lot-to-lot consistency may vary. It is advisable to validate each new lot using standardized protocols.
