Biological Background: IDO1 Function and Localization

  • IDO1 (Indoleamine 2,3-dioxygenase 1), also known as IDO or INDO, is a heme-containing enzyme that catalyzes the first and rate-limiting step of tryptophan catabolism along the kynurenine pathway.
  • It plays a crucial role in peripheral immune tolerance by depleting tryptophan, which inhibits T lymphocyte proliferation, and by producing tryptophan catabolites that promote T-cell apoptosis and regulatory T-cell differentiation. Related references: PMID:25691885
  • IDO1 acts as a suppressor of anti-tumor immunity; its expression in tumors is associated with a worse clinical outcome. Related references: PMID:14502282 PMID:23103127 PMID:25157255 PMID:18418598
  • The enzyme protects the fetus from maternal immune rejection by locally depleting tryptophan at the maternal-fetal interface. Related references: PMID:25691885
  • IDO1 also limits the growth of intracellular pathogens by depriving them of tryptophan. Related references: PMID:25691885
  • Subcellularly, IDO1 is located in the cytoplasm, specifically in the cytosol.
  • It is expressed in mature dendritic cells in lymphoid organs, some epithelial cells, and endothelial cells of the placenta, and is highly inducible in most tissues, with overexpression in over 50% of tumors.
  • Keywords: Dioxygenase, Heme, Iron, Metal-binding, Oxidoreductase, Tryptophan catabolism, Immunity.

Experimental Guidance and Technical Tips

  • This antibody is recommended for Western blotting (WB), immunofluorescence/immunocytochemistry (IF/ICC), and ELISA applications. For WB, consider optimizing loading amounts based on the expected molecular weight (~45 kDa).
  • Given the immunogen design covering the full-length protein (amino acids 1-403), this antibody may recognize both native and denatured forms of IDO1 in human samples.
  • Since cross-reactivity is validated only for human, ensure validation of specificity in your relevant experimental system, especially when using non-human samples.
  • Perform pilot experiments to optimize antibody dilutions and incubation conditions for your specific application and sample type.
