Biological Background: IDO1 Function and Localization
- IDO1 catalyzes the first and rate-limiting step of tryptophan catabolism along the kynurenine pathway. Related references: PMID:17671174 PMID:18026683
- It plays a critical role in peripheral immune tolerance by depleting tryptophan, which inhibits T lymphocyte division, and by accumulating tryptophan catabolites that induce T-cell apoptosis and differentiation of regulatory T-cells. Related references: PMID:25691885
- IDO1 acts as a suppressor of anti-tumor immunity, contributing to tumor immune escape. Related references: PMID:14502282 PMID:23103127 PMID:25157255
- It limits the growth of intracellular pathogens by depriving them of tryptophan. Related references: PMID:25691885
- The enzyme protects the fetus from maternal immune rejection during pregnancy. Related references: PMID:25691885
- IDO1 is primarily localized in the cytoplasm and cytosol.
- Tissue expression is observed in mature dendritic cells in lymphoid organs, some epithelial cells, endothelial cells of placenta, and is inducible in most tissues; it is overexpressed in over 50% of tumors, correlating with poor clinical outcome.
- IDO1 is a heme-containing dioxygenase that binds iron, belonging to the tryptophan catabolism pathway.
Experimental Guidance and Technical Tips
- For Western blot, an expected band around 45 kDa may be observed; consider loading appropriate positive controls or using lysates from IDO1-expressing tissues.
- In immunofluorescence (IF/ICC), optimize fixation and permeabilization conditions as IDO1 is a cytosolic protein.
- Validate antibody performance in the relevant sample system, as IDO1 expression is often inducible and varies across tissues.
