Biological Background: Insulin-degrading enzyme Function and Localization
- IDE (Insulin-degrading enzyme), also known as Insulysin, Abeta-degrading protease, or Insulin protease, is a zinc-dependent metalloprotease encoded by the IDE gene.
- It plays a crucial role in degrading insulin, glucagon, and islet amyloid polypeptide (IAPP), thereby regulating blood glucose homeostasis and peptide hormone signaling (PubMed:10684867, PubMed:17051221, PubMed:17613531). Related references: PMID:10684867, PMID:17051221, PMID:17613531
- IDE degrades amyloid-beta (Aβ) peptides derived from amyloid precursor protein (APP), contributing to the clearance of neurotoxic Aβ aggregates and potentially impacting Alzheimer's disease pathology (PubMed:26394692, PubMed:9830016). Related references: PMID:26394692, PMID:9830016
- It also degrades natriuretic peptides (ANP, BNP, CNP) and an aberrant frameshifted form of NPPA (fsNPPA) linked to familial atrial fibrillation, modulating cGMP signaling (PubMed:21098034). Related references: PMID:21098034
- Subcellularly, IDE localizes to the cytoplasm, cytosol, cell membrane, and is secreted, with expression detected in brain and cerebrospinal fluid.
- The membrane-associated isoform functions as an entry receptor for varicella-zoster virus (VZV), highlighting a role in host-virus interaction.
- IDE is characterized as an allosteric enzyme with ATP-binding and metal-binding properties, and undergoes alternative splicing.
- Additionally, IDE participates in antigen processing, generating the MAGEA3-derived antigenic peptide presented by MHC class I.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-250 of human IDE; therefore, this antibody may detect full-length IDE or N-terminal fragments. Validate the specificity in your sample system.
- For Western blot, a band of approximately 118 kDa is expected for human IDE. Use appropriate protein ladders and consider positive control lysates from brain tissue or cell lines expressing IDE.
- In immunohistochemistry (IHC-P), optimize antigen retrieval conditions and use appropriate tissue sections; IDE is expressed in brain and other tissues, so include relevant positive and negative controls.
- For ELISA applications, the antibody can be considered as a capture or detection reagent; titrate to determine optimal working concentrations.
- As a [KD Validated] antibody, it may be suitable for knockdown confirmation studies; employ appropriate siRNA/shRNA controls.
