Biological Background: ID3 Function and Localization
- ID3 (inhibitor of DNA binding 3) is a transcriptional regulator that lacks a basic DNA-binding domain. It negatively regulates basic helix-loop-helix (bHLH) transcription factors by forming heterodimers, thereby inhibiting their DNA binding and transcriptional activity.
- ID3 is implicated in a broad range of cellular processes, including cellular growth, senescence, differentiation, apoptosis, angiogenesis, and neoplastic transformation.
- In myogenesis, ID3 inhibits skeletal muscle and cardiac myocyte differentiation while promoting the proliferation of muscle precursor cells. It also prevents the binding of E2A-containing protein complexes to the muscle creatine kinase E-box enhancer.
- ID3 regulates the circadian clock by repressing the transcriptional activator activity of the CLOCK-BMAL1 heterodimer.
- The protein localizes to the nucleus, consistent with its role as a transcriptional repressor.
- ID3 is abundantly expressed in lung, kidney, and adrenal gland, but not in adult brain.
