Biological Background: DNA-binding protein inhibitor ID-1 Function and Localization

  • DNA-binding protein inhibitor ID-1 (ID1), also known as inhibitor of DNA binding 1 or inhibitor of differentiation 1, is a member of the ID family of helix-loop-helix (HLH) proteins that lack a basic DNA-binding domain. Alternative names include Class B basic helix-loop-helix protein 24 (bHLHb24).
  • ID1 functions as a transcriptional regulator by negatively regulating basic helix-loop-helix (bHLH) transcription factors. It forms heterodimers with bHLH proteins, inhibiting their DNA binding and transcriptional activity.
  • ID1 is implicated in a variety of cellular processes, including cellular growth, senescence, differentiation, apoptosis, angiogenesis, and neoplastic transformation. It inhibits skeletal muscle and cardiac myocyte differentiation.
  • ID1 regulates the circadian clock by repressing the transcriptional activator activity of the CLOCK-BMAL1 heterodimer (by similarity).
  • Subcellular localization: ID1 is found in both the cytoplasm and nucleus, consistent with its role in nucleocytoplasmic shuttling and transcriptional regulation.
  • UniProt keywords associated with ID1 include alternative splicing, biological rhythms, developmental protein, nucleus, repressor, and transcription regulation.

Experimental Guidance and Technical Tips

  • For western blotting, the predicted molecular weight of ID1 is 16 kDa; use appropriate gel percentage and transfer conditions for low molecular weight proteins.
  • ID1 may be expressed at low levels in certain tissues or cell lines; consider optimizing sample loading and detection sensitivity.
  • The immunogen corresponds to full-length human ID1; cross-reactivity with mouse and rat is expected based on homology, but validation in the relevant species' sample system is recommended.
  • For ELISA, empirical optimization of coating antigen concentration and detection conditions is advised.
  • Given ID1's ability to shuttle between cytoplasm and nucleus, subcellular fractionation may aid in studying its localization-specific functions.
