Biological Background: Inducible T-cell costimulator Function and Localization
- ICOS (Inducible T-cell costimulator), also known as AILIM, CCLP, CRP-1, is a stimulatory receptor expressed on activated T-cells and resting memory T-cells.
- Upon binding to its ligand ICOSL on antigen-presenting cells, ICOS delivers costimulatory signals that enhance T-cell proliferation, lymphokine secretion (including IL-10), up-regulation of cell-cell interaction molecules, and effective help for B-cell antibody secretion. - ICOS is critical for the differentiation of T follicular regulatory cells upon immune challenges such as viral infection. Related references: PMID:36754569
- Mechanistically, ICOS potentiates TCR-induced calcium flux by augmenting PLCG1 activation and actin remodeling. Related references: PMID:27693916
- In addition, ICOS activates PI3K signaling pathways independently of calcium flux. Related references: PMID:19915142, PMID:27693916
- ICOS plays a critical role in CD40-mediated class switching of immunoglobin isotypes. Related references: PMID:11343122
- ICOS also prevents the apoptosis of pre-activated T-cells and is essential for efficient interaction between T and B-cells and for normal antibody responses to T-cell dependent antigens.
- Subcellularly, ICOS localizes to the cell membrane and is expressed on activated T-cells, resting memory T-cells, high in thymic medulla, germinal centers, and T-cell zones of lymph nodes and Peyer patches.
- As a glycoprotein, ICOS contains an immunoglobulin domain, disulfide bonds, and a transmembrane helix, consistent with its receptor function.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular region of mouse ICOS, making this antibody suitable for detecting native cell surface ICOS by flow cytometry (FC) and immunofluorescence (IF/ICC).
- For Western blot (WB), the predicted molecular weight is approximately 22 kDa; consider using reducing conditions and appropriate membrane blocking. Validate antibody performance in your specific sample system, as expression may vary with T-cell activation status.
- This antibody is raised against mouse ICOS and shows reactivity with mouse; cross-reactivity with other species has not been validated. For human or other species, confirm cross-reactivity experimentally.
