Biological Background: ICK Function and Localization
- ICK (serine/threonine-protein kinase ICK), also known as CILK1, hICK, LCK2, and MRK, is a member of the MAK-related kinase family essential for ciliogenesis and developmental signaling.
- It is required for primary cilium formation; knockout or mutation leads to defective ciliogenesis. Related references: PMID:24797473
- ICK phosphorylates the kinesin motor protein KIF3A, a key component of intraflagellar transport (IFT) required for ciliary assembly and function.
- The kinase controls ciliary length by modulating IFT speed in a cAMP- and mTORC1-dependent manner. Related references: PMID:24853502
- ICK localizes to multiple subcellular compartments including the nucleus, cytoplasm, cytosol, and specifically to the cilium and ciliary basal body, underscoring its role in ciliary dynamics.
- Tissue expression is broad, with highest levels in placenta and testis, and expression in heart, brain, pancreas, thymus, prostate, ovary, and intestinal tissues; it is absent from spleen and is frequently detected in cancer cell lines.
- The protein is subject to alternative splicing and is classified as an ATP-binding, magnesium-dependent kinase involved in developmental processes and associated with ciliopathies, craniosynostosis, ectodermal dysplasia, and epilepsy.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 1–270 of human ICK (NP_057597.2), encompassing the kinase domain; this region may contribute to broad species cross-reactivity (human, mouse, rat) as validated by the supplier.
- For Western blot: consider using reducing conditions and probing lysates from ciliated cell lines or tissues known to express ICK (e.g., testis, placenta, or cancer lines). An ~71 kDa band is expected.
- As a polyclonal antibody, lot-to-lot consistency should be verified; include positive and negative controls (e.g., ICK knockout or siRNA-treated samples) to confirm specificity in your experimental system.
- ELISA-based detection may require pairing with a suitable capture antibody or optimized coating conditions; pilot experiments are recommended.
