Biological Background: ICAM3/CD50 Function and Localization
- ICAM3 (CD50) is a type I transmembrane glycoprotein of the immunoglobulin superfamily, also known as CDw50 and ICAM-R.
- ICAM3 functions as a cell adhesion molecule by serving as a ligand for the integrin LFA-1 (ITGAL/ITGB2), mediating leukocyte adhesion, signaling, and the phagocytosis of apoptotic neutrophils by macrophages. Related references: PMID:1448173 PMID:1448174 PMID:23775590
- It also acts as a ligand for the integrin ITGAD/ITGB2, expanding its role in immune cell interactions. Related references: PMID:8777714
- ICAM3 binds dendritic cell-specific ICAM-3-grabbing non-integrin (CD209) in a Ca2+-dependent manner, facilitating initial DC–T cell contacts, clustering, and T cell proliferation. Related references: PMID:10721994
- The protein is constitutively expressed on the surface of resting leukocytes but is absent from resting or cytokine-activated endothelial cells. Related references: PMID:8325327 PMID:1448174
- ICAM3 undergoes phosphorylation and glycosylation; its immunoglobulin-like domains are stabilized by disulfide bonds.
- The predicted molecular mass is approximately 59.5 kDa, with glycosylation resulting in an apparent weight of 60–85 kDa on SDS-PAGE.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal extracellular region (aa 30–114) of human ICAM3, encompassing the ligand-binding domain; this design may favor recognition of functionally relevant epitopes.
- For western blotting, validate detection in leukocyte lysates; expect a band around 60–85 kDa due to variable glycosylation.
- For immunohistochemistry, consider antigen retrieval and include appropriate positive controls such as tonsil or lymph node tissues.
- In flow cytometry, use live, non-permeabilized leukocytes to detect surface expression; titrate the antibody for optimal signal-to-noise.
- For immunofluorescence and immunocytochemistry, fix and permeabilize cells appropriately, and confirm localization at the plasma membrane.
