Biological Background: ICAM3 Function and Localization
- ICAM3 (Intercellular adhesion molecule 3), also designated CD50 or ICAM-R, is a single-pass type I membrane protein and member of the immunoglobulin superfamily, characterized by multiple immunoglobulin-like domains and intramolecular disulfide bonds.
- It serves as a primary ligand for the integrin LFA-1 (ITGAL:ITGB2), playing a crucial role in immune cell adhesion and the phagocytosis of apoptotic neutrophils by macrophages. Related references: PMID:1448173 PMID:1448174 PMID:23775590
- Additionally, ICAM3 interacts with integrin ITGAD/ITGB2, expanding its repertoire of leukocyte adhesion functions. Related references: PMID:8777714
- Through a Ca2+-dependent, integrin-independent mechanism, ICAM3 binds CD209 on dendritic cells, initiating dendritic cell–T cell clustering and facilitating T cell proliferation. Related references: PMID:10721994

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 270–490 within the extracellular region of human ICAM3, which includes domains likely involved in ligand binding. This design may elicit antibodies recognizing native epitopes; however, antibody specificity should be confirmed using appropriate controls (e.g., knockout or knockdown cell lines).
- For Western blot (WB) analysis, it is recommended to use protein lysates from human leukocyte cell lines or primary leukocytes, where ICAM3 is abundantly expressed. A band at approximately 60 kDa is expected; consider using a molecular weight marker and a known positive control.
- In ELISA applications, the polyclonal nature of this antibody allows flexible pairing (e.g., as capture or detection antibody) when combined with a suitable monoclonal partner. Validate the optimal dilution and buffer conditions in the target assay system.
- Note that this antibody is specific for human ICAM3; cross-reactivity with mouse or rat orthologs is unlikely based on limited sequence homology in the immunogen region. If extending to other species, verify sequence alignment and test experimentally.
