Biological Background: ICAM2/CD102 Function and Localization
- Encoded by the Icam2 gene, intercellular adhesion molecule 2 (ICAM2) is a type I transmembrane glycoprotein belonging to the immunoglobulin superfamily; also known as CD102 or Ly-60 in mouse.
- Functions as a cell adhesion molecule, serving as a receptor ligand for the integrin ITGAL:ITGB2 (LFA-1) on leukocytes, mediating cell-cell adhesion on resting cells. Related references: PMID:1401904
- Additionally, acts as a receptor for the dendritic cell (DC) lectin CD209, triggering transendothelial migration of DC precursors from blood into peripheral and lymphoid tissues.
- Localizes to the cell membrane and is enriched on microvilli of endothelial cells and leukocytes, facilitating rapid tethering and adhesion.
- Highly expressed in lung tissue, with constitutive presence in endothelial cells and leukocytes, playing a role in lymphocyte recirculation and immune surveillance.
- Contains two immunoglobulin-like C2-type domains in the extracellular region, a single-pass transmembrane helix, and a short cytoplasmic tail; the mature protein has a calculated molecular mass of approximately 31.4 kDa.

Experimental Guidance and Technical Tips
- The immunogen used was a recombinant protein corresponding to the extracellular domain (amino acids 20-222). Therefore, this antibody is expected to recognize the extracellular region of mouse ICAM2.
- For Western blotting, a band at approximately 31 kDa should be observed. It is advisable to include appropriate positive controls (e.g., mouse lung lysate) and validate under reducing/non-reducing conditions.
- In immunohistochemistry (IHC-P), antigen retrieval may be required; mouse lung sections serve as a suitable positive control given the high expression in this tissue.
- For ELISA applications, consider pairing with another clone or using a recombinant protein standard to establish assay conditions; the antibody can be used for either capture or detection.
- As a rabbit-derived recombinant monoclonal antibody (PolymAb®), it exhibits high specificity and batch-to-batch consistency. Use an anti-rabbit secondary antibody for detection.
