Biological Background: ICAM-1/CD54 Function and Localization
- ICAM-1 (Intercellular adhesion molecule 1), also known as CD54 or Major group rhinovirus receptor, is a cell adhesion molecule encoded by the ICAM1 gene (GeneID: 3383). It is a single-pass type I membrane protein belonging to the immunoglobulin superfamily.
- ICAM-1 serves as a ligand for the integrin LFA-1 (ITGAL:ITGB2), mediating leukocyte cell-cell adhesion and contributing to calibrated T-cell killing responses. Related references: PMID:3086451, PMID:3340213, PMID:38195629
- It also binds to MAC-1 (ITGAM:ITGB2) on stimulated neutrophils, promoting adhesion to endothelial cells. Related references: PMID:1980124
- During leukocyte trans-endothelial migration, ICAM-1 engagement triggers assembly of endothelial apical cups via ARHGEF26/SGEF and RHOG activation. Related references: PMID:17875742
- As a pathogen receptor, ICAM-1 acts as the entry receptor for major group rhinoviruses A and B and Coxsackievirus A21.
- Upon Kaposi's sarcoma-associated herpesvirus/HHV-8 infection, ICAM-1 is targeted for degradation by the viral E3 ubiquitin ligase MIR2, likely to evade immune detection.
- ICAM-1 is located on the cell membrane, with expression on non-hematopoietic cells (vascular endothelial cells, epithelial cells, fibroblasts) and hematopoietic cells (tissue macrophages, T-cell blasts, dendritic cells), and at low levels on peripheral blood leukocytes. Related references: PMID:3086451
- Key post-translational modifications include glycosylation, disulfide bond formation, and phosphorylation. The protein contains immunoglobulin-like domains and a transmembrane helix.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain of human ICAM-1 (amino acids 250-480), which may be suitable for detecting surface-expressed or soluble protein.
- For Western blotting, consider a predicted molecular weight of approximately 58 kDa; however, glycosylation and other modifications may shift the apparent molecular weight.
- Validate antibody reactivity in the relevant sample system, particularly when using mouse samples, due to cross-reactivity claims.
- For ELISA, test against recombinant ICAM-1 protein or cell lysates to determine optimal working dilutions.
