Biological Background: IAPP Function and Localization
- Islet amyloid polypeptide (IAPP), also known as amylin or diabetes-associated peptide (DAP), is a glucoregulatory peptide hormone with a molecular weight of approximately 9.8 kDa.
- IAPP selectively inhibits insulin-stimulated glucose utilization and glycogen deposition in skeletal muscle, while sparing adipocyte glucose metabolism, thereby contributing to energy homeostasis. Related references: PMID:2690069
- Signaling occurs through calcitonin receptor (CALCR) and receptor activity-modifying protein (RAMP) complexes, forming high-affinity amylin receptors (AMYRs); IAPP can also bind CALCR alone with lower affinity (By similarity).
- The hormone is secreted from pancreatic islet β-cells and co-released with insulin in response to nutrient stimuli.
- Post-translational processing involves signal peptide removal, cleavage at paired basic residues to generate the 37-amino acid mature peptide, C-terminal amidation, and intrachain disulfide bond formation.
- IAPP is intrinsically amyloidogenic; its aggregation into amyloid deposits in pancreatic islets is a pathological hallmark of type 2 diabetes.
- UniProt keywords include: 3D-structure, Amidation, Amyloid, Cleavage on pair of basic residues, Disulfide bond, Hormone, Secreted, and Signal.
Experimental Guidance and Technical Tips
- The immunogen encompasses the full-length human IAPP precursor (aa 1–89); the antibody may recognize both pro-IAPP and mature IAPP. Validate expected band sizes (~10 kDa for precursor, ~4 kDa for mature peptide) under reducing and non-reducing conditions.
- For Western blotting, a predominant band near 10 kDa is anticipated; use low molecular weight markers and optimize transfer conditions to retain small peptides.
- In immunohistochemistry (paraffin-embedded, IHC-P) and immunofluorescence (IF-P), employ antigen retrieval and include appropriate positive/negative controls, particularly in pancreatic or brain tissues where IAPP is expressed.
- ELISA applications can leverage the polyclonal's broad epitope coverage; consider pairing with a monoclonal capture antibody for sandwich assays to improve signal-to-noise ratio.
- Reported cross-reactivity with mouse and rat IAPP should be independently validated in the relevant sample system due to interspecies sequence differences.
