Biological Background: HYAL1 Function and Localization
- HYAL1, also known as hyaluronoglucosaminidase-1 or LuCa‑1, is a glycosidase/hydrolase enzyme that belongs to the hyaluronidase protein family.
- The protein contains an EGF-like domain, multiple disulfide bonds, and features both N-glycosylation and a signal peptide.
- Predominantly localized to lysosomes and also secreted, HYAL1 may contribute to tumor progression by modulating the extracellular matrix.
- It may negatively regulate TGF‑β1‑mediated cell proliferation, suggesting a context‑dependent role in growth signaling.
- HYAL1 is highly expressed in liver, kidney, and heart, with lower levels in lung, placenta, and skeletal muscle, and undetectable in adult brain. Specific isoforms are selectively expressed in bladder and prostate cancers.
- Mutations in HYAL1 are associated with mucopolysaccharidosis type IX, a lysosomal storage disorder.
- Multiple isoforms are generated by alternative splicing, and the protein has been confirmed by direct protein sequencing and proteomics identification.
Experimental Guidance and Technical Tips
- The immunogen (aa 22‑435) covers most of the mature HYAL1 sequence, making it likely to recognize multiple isoforms and post-translationally modified forms.
- For Western blot applications, expect bands between 10 and 48 kDa due to alternative splicing; validate the observed molecular weights against the specific isoform(s) expressed in your sample type.
- In ELISA, this monoclonal antibody can be used for capture/detection of HYAL1; however, optimal dilutions and assay conditions must be determined by the end user.
- Cross-reactivity with non-human species has not been tested; verify reactivity if using samples from other organisms.
