Biological Background: HYAL1 Function and Localization

• HYAL1, also known as hyaluronoglucosaminidase-1 or LuCa‑1, is a glycosidase/hydrolase enzyme that belongs to the hyaluronidase protein family.
• The protein contains an EGF-like domain, multiple disulfide bonds, and features both N-glycosylation and a signal peptide.
• Predominantly localized to lysosomes and also secreted, HYAL1 may contribute to tumor progression by modulating the extracellular matrix.
• It may negatively regulate TGF‑β1‑mediated cell proliferation, suggesting a context‑dependent role in growth signaling.
• HYAL1 is highly expressed in liver, kidney, and heart, with lower levels in lung, placenta, and skeletal muscle, and undetectable in adult brain. Specific isoforms are selectively expressed in bladder and prostate cancers.
