Biological Background: CD8a Function and Localization
- T-cell surface glycoprotein CD8 alpha chain (alternative name: T-lymphocyte differentiation antigen T8/Leu-2) is a single-pass type I membrane protein encoded by the CD8A gene.
- Functions primarily as a coreceptor for MHC class I molecules, interacting simultaneously with the T-cell receptor (TCR) and MHC class I:peptide complexes on antigen-presenting cells.
- Recruits the Src kinase LCK to the TCR-CD3 complex, initiating intracellular signaling pathways that lead to lymphokine production, motility, adhesion, and activation of cytotoxic T-lymphocytes (CTLs).
- Enables CTLs to recognize and eliminate infected cells and tumor cells; in NK cells, CD8A homodimers promote conjugation and lysis of multiple target cells and support survival and differentiation into memory CD8 T-cells.
- Localizes to the cell membrane and can be secreted; expressed as a disulfide-linked alpha/beta heterodimer on thymus-derived T-cells or as a CD8A homodimer on NK cells, memory T-cells, intraepithelial lymphocytes, monocytes, and dendritic cells.
- Contains an immunoglobulin-like domain, is glycosylated, palmitoylated, and forms disulfide bonds; molecular weight varies (21-30 kDa) due to alternative splicing and post-translational modifications.
