  • CD8A is a ~25.7 kDa type I transmembrane glycoprotein, also known as Leu-2 or T8/Leu-2, belonging to the immunoglobulin superfamily. Multiple isoforms exist due to alternative splicing.
  • Functions as a co-receptor for the T-cell receptor (TCR) by binding to MHC class I molecules on antigen-presenting cells, critical for thymic T-cell selection and peripheral immune surveillance.
  • Upon TCR engagement, the cytoplasmic tail of CD8A associates with the Src-family kinase LCK, initiating signaling cascades that drive cytokine production, cytotoxicity, and T-cell proliferation.
  • In cytotoxic T-lymphocytes (CTLs), CD8A-MHC class I interaction enables recognition and elimination of virus-infected cells and tumor cells.
  • CD8A can also form homodimers on natural killer (NK) cells, promoting conjugate formation, target cell lysis, and survival signaling.
  • Expressed predominantly on the surface of CD8+ T-cells as a heterodimer with CD8B; homodimers are found on subsets of NK cells, memory T-cells, intraepithelial lymphocytes, monocytes, and dendritic cells.
  • Subcellular location: primarily plasma membrane; also secreted as a soluble form following activation.
