Biological Background: CD69 Function and Localization
- CD69 (Cluster of Differentiation 69), also known as AIM, EA1, MLR-3, CLEC2C, GP32/28, and BL-AC/P26, is a type II transmembrane glycoprotein belonging to the C-type lectin family.
- It functions as an early activation marker rapidly expressed on the surface of platelets, T-lymphocytes, and NK cells upon stimulation, playing a central role in immune cell homeostasis.
- CD69 negatively regulates Th17 cell differentiation by interacting with galectin-1 on immature dendritic cells, thereby modulating adaptive immunity. Related references: PMID:24752896
- It promotes the conversion of naive CD4+ T-cells into regulatory T-cells via the S100A8/S100A9 complex, contributing to immune tolerance. Related references: PMID:26296369
- Acting as an oxidized low-density lipoprotein (oxLDL) receptor in CD4+ T-lymphocytes, CD69 negatively regulates inflammation by inducing PDCD1 expression through NFAT activation. Related references: PMID:35930205
- It participates in adipose tissue-derived mesenchymal stem cell-mediated defense against Pseudomonas aeruginosa infection by promoting ERK1 activation and GM-CSF secretion. Related references: PMID:34841721
- CD69 acts as a S1PR1 agonist, facilitating receptor internalization and degradation to negatively regulate chemotactic responses of effector lymphocytes and dendritic cells to sphingosine 1-phosphate. Related references: PMID:37039481
- The protein is predominantly localized to the cell membrane and is expressed on a variety of activated hematopoietic cells, including T-cells, B-cells, NK cells, neutrophils, eosinophils, and platelets.
Experimental Guidance and Technical Tips
- This antibody is directly conjugated to , eliminating the need for secondary staining steps and reducing assay complexity in flow cytometry.
- Owing to its specificity for an extracellular epitope (amino acids 78–199), it is suitable for surface staining of live, unfixed cells.
- Consider titrating the antibody to determine the optimal concentration for your specific cell type and activation conditions; use appropriate isotype controls for accurate gating.
- The documented cross-reactivity with cynomolgus monkey (Macaca fascicularis) makes this reagent suitable for non-human primate models in preclinical studies.
- When designing multicolor panels, ensure spectral compatibility with other fluorophores to avoid compensation issues.
