Biological Background: TNFRSF11A Function and Localization
- Tumor necrosis factor receptor superfamily member 11A (TNFRSF11A), also known as RANK, ODFR, and receptor activator of NF-κB, belongs to the TNF receptor family.
- Receptor for TNFSF11/RANKL (TRANCE/OPGL); essential for RANKL-mediated osteoclastogenesis. Related references: PMID:9878548
- Mediates signaling through interactions with EEIG1, promoting NFATC1 transcription and PLCG2 activation.
- Plays a role in regulating interactions between T-cells and dendritic cells.
- Localized at the cell membrane and membrane rafts.
- Ubiquitously expressed with highest levels in skeletal muscle, thymus, liver, colon, small intestine, and adrenal gland.
- Undergoes alternative splicing; carries disulfide bonds, glycosylation, and phosphorylation; features a signal peptide, transmembrane domain, and extracellular cysteine-rich repeats.
- Associated with diseases including osteopetrosis and deafness.
Experimental Guidance and Technical Tips
- For WB, observe a band around 66 kDa (predicted molecular weight) in human, mouse, and rat samples; validate using appropriate positive control tissues such as thymus or skeletal muscle.
- This polyclonal antibody is suitable for ELISA; consider using the recombinant immunogen (amino acids 30–212) as a positive control to optimize assay conditions.
- Due to post-translational modifications (glycosylation, phosphorylation), the observed molecular weight may differ slightly from the predicted 66 kDa; treat samples with phosphatase or deglycosylating enzymes if necessary.
- For cross-reactive studies in mouse and rat, confirm sequence homology in the immunogen region to ensure expected reactivity; the immunogen aligns with human sequence, but cross-reactivity has been supplier-validated.
- Consider blocking endogenous biotin or peroxidases when using sensitive detection systems in tissues with high metabolic activity.
