Biological Background: TIM-3/HAVCR2 Function and Localization
-
TIM-3 (HAVCR2, CD366) is a cell surface receptor that generally inhibits innate and adaptive immune responses, and regulates macrophage activation; context-dependent stimulating functions have also been reported.

Inhibits T-helper type 1 (Th1)-mediated auto- and alloimmune responses and promotes immunological tolerance.

In CD8+ T-cells, attenuates TCR-induced signaling by blocking NF-kappaB and NFAT activities, reducing IL-2 secretion.

Expressed on Treg cells and inhibits Th17 cell responses.

Functions as a receptor for LGALS9 (galectin-9), leading to T-cell suppression and apoptosis; however, this receptor function has been challenged.

On natural killer (NK) cells, enhances IFN-gamma production but can also suppress cytotoxicity.

Localizes at the cell membrane and cell junctions; expressed in Th1, Treg, dendritic cells, NK cells, and epithelial tissues, with upregulation in chronic HCV and HIV infections.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 22–200) of human TIM-3, potentially enabling recognition of the native receptor in flow cytometry.
- This antibody is validated for flow cytometry (FC). For other applications (e.g., Western blot, IHC), consider empirical optimization and validation in the relevant sample system.
- Given TIM-3 expression on multiple immune cell types, include appropriate gating strategies and cell-type markers when analyzing complex cell populations.
