Biological Background: Thioredoxin Reductase 1 Function and Localization
- Thioredoxin reductase 1 (TXNRD1), also known as TR1, TXNR, GRIM-12, or KM-102-derived reductase-like factor, is a homodimeric flavoprotein encoded by the TXNRD1 gene. It belongs to the thioredoxin reductase family and contains a selenocysteine residue at its C‑terminal active site, essential for catalysis.
- TXNRD1 catalyzes the reduction of thioredoxin (Trx) from its disulfide to dithiol form, a central reaction in the regulation of cellular redox homeostasis, growth, and differentiation (PubMed:8577704). Related references: PMID:8577704
- The enzyme also exhibits peroxidase activity, directly reducing hydrogen peroxide (H2O2) (PubMed:10849437). Related references: PMID:10849437
- TXNRD1 induces actin and tubulin polymerization, leading to the formation of cell membrane protrusions.
- It enhances the transcriptional activity of estrogen receptors ESR1 and ESR2, with specific enhancement noted for ESR2 (PubMed:15199063). Related references: PMID:15199063
- The protein mediates cell death induced by a combination of interferon‑beta and retinoic acid (PubMed:9774665). Related references: PMID:9774665
- TXNRD1 is predominantly localized in the cytoplasm, with additional localization observed in the nucleus.
- At the protein level, TXNRD1 is expressed predominantly in Leydig cells and is also detected in ovary, spleen, heart, liver, kidney, pancreas, and various cancer cell lines. It shows widespread tissue distribution, with highest mRNA levels in kidney, testis, uterus, ovary, prostate, placenta, and fetal liver.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering the full-length human TXNRD1 (amino acids 1–649); consider verifying detection of the endogenous protein in your experimental context, as epitope accessibility may vary.
- For Western blotting, reduced denaturing conditions are typically suitable; expect a band at approximately 71 kDa. Validate with appropriate positive controls (e.g., HeLa, HepG2, or testis lysates) for your specific sample type.
- This rabbit monoclonal antibody is recommended for ELISA and WB. Cross‑reactivity with mouse and rat TXNRD1 has been reported; however, independent validation in your tissue or cell line is advised before application.
- Given TXNRD1's central role in redox control, estrogen signaling, and its overexpression in many cancers, consider testing this antibody in models of oxidative stress or hormone‑dependent tumorigenesis.
