Biological Background: SYT12 Function and Localization
- Synaptotagmin-12 (SYT12), also known as Synaptotagmin XII (SytXII), is a member of the synaptotagmin family of synaptic vesicle proteins.
- Unlike many other synaptotagmins, SYT12 does not bind Ca²⁺ or phospholipids, suggesting a distinct regulatory mechanism in neurotransmitter release.
- SYT12 enhances spontaneous neurotransmitter release but does not affect induced neurotransmitter release, indicating a role in basal synaptic activity.
- Essential for mossy-fiber long-term potentiation (LTP) in the hippocampus, a process critical for learning and memory.
- Localized to the synaptic vesicle membrane within cytoplasmic secretory vesicles, consistent with its function in synaptic transmission.
- Identified as a phosphoprotein, potentially subject to activity-dependent regulation.
- Contains transmembrane helices and repeat regions, characteristic of synaptotagmin family topology.
- The gene SYT12 encodes a protein with a calculated molecular mass of 46.5 kDa (UniProt), though observed at 47 kDa under experimental conditions.
Experimental Guidance and Technical Tips
- The immunogen is designed against amino acids 95–200 of human SYT12, an internal region; thus, the antibody is expected to detect full-length SYT12.
- For Western Blot (WB), consider using human or mouse brain tissue lysates as positive controls, given SYT12’s expression in the central nervous system and the observed cross-reactivity with mouse.
- For Immunofluorescence/Immunocytochemistry (IF/ICC), optimize fixation and permeabilization conditions to preserve synaptic vesicle membrane epitopes.
- In ELISA applications, the recombinant immunogen or native SYT12 can be used for antibody titration and specificity assessment.
- No cross-reactivity data is available for species other than human and mouse; validate reactivity in target species if required.
