Biological Background: SIGLEC9 Function and Localization

  • SIGLEC9 (Sialic acid-binding Ig-like lectin 9) is also known as CD329, CDw329, or protein FOAP-9. It belongs to the immunoglobulin superfamily and sialic acid-binding lectin (siglec) family.
  • Functionally, SIGLEC9 acts as a putative adhesion molecule that mediates sialic-acid-dependent binding to cells. It preferentially binds alpha-2,3- or alpha-2,6-linked sialic acids.
  • The sialic acid recognition site of SIGLEC9 may be masked through cis interactions with sialic acids on the same cell surface, which can regulate its binding activity.
  • SIGLEC9 is a single-pass type I membrane protein, localized to the plasma membrane.
  • Tissue expression is highest in peripheral blood leukocytes, specifically neutrophils and monocytes, and also detected in liver, fetal liver, bone marrow, placenta, and spleen. Lower levels are found in various other tissues.
  • The protein contains two immunoglobulin-like domains (one V-set and one C2-set), a transmembrane helix, and a cytoplasmic tail. It is subject to alternative splicing and is glycosylated.
  • Key post-translational modifications include disulfide bonds and N-linked glycosylation. The molecular weight of the canonical isoform is around 50 kDa, with additional isoforms potentially detected at ~52 kDa.

  • The immunogen comprises amino acids 18–348 of human SIGLEC9, covering the entire extracellular domain. This design typically generates antibodies that recognize native membrane-bound SIGLEC9.
  • For flow cytometry, titrate the antibody to determine the optimal concentration for your sample type. Include appropriate isotype controls and live/dead staining to ensure specificity.
  • Because SIGLEC9 can engage in cis interactions with cell-surface sialic acids, the antibody epitope may be partially masked on intact cells. If weak staining is observed, consider treating cells with neuraminidase (sialidase) or using acid wash conditions to disrupt cis-binding and improve antibody access.
  • The conjugate provides a bright and photostable signal suitable for multi-color flow cytometry panels. Avoid repeated freeze-thaw cycles and protect from light to maintain conjugate integrity.
