Biological Background: ROR1 Function and Localization

  • ROR1 is an inactive tyrosine-protein kinase transmembrane receptor (also designated Neurotrophic tyrosine kinase, receptor-related 1). It exhibits very low kinase activity in vitro and is unlikely to function as a tyrosine kinase in vivo. Related references: PMID:25029443
  • It functions as a receptor for WNT5A, activating downstream NFκB signaling and possibly inhibiting WNT3A-mediated pathways. Related references: PMID:25029443, PMID:27162350
  • In the inner ear, ROR1 is essential for the innervation of auditory hair cells by spiral ganglion neurons. Related references: PMID:27162350
  • Through binding to IGFBP5, ROR1 forms a complex with ERBB2 that enhances CREB oncogenic signaling. Related references: PMID:36949068
  • The receptor localizes to the cell membrane and cell projections such as axons.
  • ROR1 is strongly expressed in heart, lung, and kidney, but only weakly in the adult CNS. The short isoform is strongly expressed in fetal and adult CNS as well as in various cancers.
  • The protein contains immunoglobulin and kringle domains and is classified as a membrane glycoprotein involved in the Wnt signaling pathway.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (amino acids 24-403), making the antibody suitable for detecting native receptor on the cell surface in flow cytometry.
  • For FC, consider using appropriate isotype controls, live/dead discrimination, and careful titration to minimize non-specific binding.
  • Fixation and permeabilization may be required for intracellular detection; however, the fluorophore is compatible with standard protocols if validated in your system.
  • Validate the antibody in a relevant human cell line or tissue type known to express ROR1 (e.g., cancer cells, fetal CNS).
