Biological Background: MMP9 Function and Localization
- Matrix metalloproteinase-9 (MMP9), also known as gelatinase B, is a secreted zinc-dependent endopeptidase belonging to the matrix metalloproteinase family. It plays essential roles in extracellular matrix (ECM) remodeling and cell migration.
- Critical for local proteolysis of the ECM and leukocyte migration, facilitating immune cell trafficking through tissues. Related references: PMID:12879005 PMID:1480034 PMID:2551898
- Cleaves a broad range of substrates, including type IV and V collagens, fibronectin, KiSS1, and NINJ1, generating biologically active fragments. Related references: PMID:1480034 PMID:12879005 PMID:32883094
- Localized to the extracellular space and extracellular matrix, consistent with its function in pericellular proteolysis.
- Detected in neutrophils at the protein level and produced by normal alveolar macrophages and granulocytes. Related references: PMID:7683678
- Contains conserved structural domains, including a zinc-binding catalytic site, and requires calcium for stability and activity. Synthesized as an inactive zymogen requiring proteolytic activation.
- Post-translationally modified by glycosylation and disulfide bond formation, which are critical for its proper folding and secretion.
Experimental Guidance and Technical Tips
- For intracellular flow cytometry, ensure thorough fixation and permeabilization using validated protocols. Test staining with known MMP9-positive human cell types, such as neutrophils or macrophage cell lines.
- The immunogen spans nearly the full-length human MMP9 (aa 20–707), providing broad epitope coverage. Confirm specificity by parallel analysis of MMP9-knockout or -knockdown cellular models when available.
- Given MMP9’s role in leukocyte migration, consider evaluating expression in stimulated versus resting immune cells to assess functional modulation.
