Biological Background: MMP9 Function and Localization
- MMP9, also known as gelatinase B or 92 kDa type IV collagenase, is a zinc-dependent endopeptidase belonging to the matrix metalloproteinase (MMP) family.
- It plays an essential role in the local proteolysis of extracellular matrix components and in leukocyte migration. Related references: PMID:12879005, PMID:1480034, PMID:2551898
- MMP9 specifically cleaves KiSS1 at a Gly-|-Leu bond. Related references: PMID:12879005
- Cleaves NINJ1 to generate the Secreted ninjurin-1 form. Related references: PMID:32883094
- Cleaves type IV and type V collagen into large C-terminal three quarter fragments and shorter N-terminal one quarter fragments. Related references: PMID:1480034
- Degrades fibronectin but not laminin or Pz-peptide.
- May play a role in bone osteoclastic resorption (By similarity).
- Detected in neutrophils and produced by normal alveolar macrophages and granulocytes; localized to the extracellular space and matrix.
- Contains calcium and zinc binding sites, disulfide bonds, and undergoes proteolytic activation from the zymogen form.
Experimental Guidance and Technical Tips
- The immunogen spans the majority of the mature MMP9 sequence (amino acids 20–707), suggesting the antibody may recognize both pro- and active forms of the enzyme. For intracellular flow cytometry, use appropriate permeabilization buffers to ensure antibody access to the intracellular compartment.
- Since cross-reactivity is listed only for human MMP9, validate specificity in your sample system of interest, especially when working with closely related MMP family members.
- For optimal results, titrate the antibody concentration and include appropriate isotype controls in your flow cytometry experiments.
- This product is recommended for research use and its performance in diagnostic applications should be independently validated.
