Biological Background: MICA/MICB Function and Localization
- MICA (MHC class I polypeptide-related sequence A) is a stress-induced ligand for the activating receptor NKG2D (KLRK1) expressed on NK cells, γδ T cells, and CD8+ αβ T cells. Related references: PMID:11491531, PMID:11777960
- Upon cellular stress such as viral/bacterial infection or DNA damage, MICA is upregulated and serves as a danger signal, triggering NK cell-mediated cytotoxicity and cytokine production. Related references: PMID:10426993
- MICA is widely expressed except in the central nervous system, with high levels in gastric epithelium, monocytes, keratinocytes, endothelial cells, and fibroblasts; it is also overexpressed in many epithelial tumors.
- Subcellularly, MICA localizes to the cell membrane and cytoplasm.
- The protein is glycosylated, palmitoylated, and can undergo alternative splicing; it contains an immunoglobulin-like domain and a transmembrane region.
