Biological Background: LILRB4 Function and Localization
- Leukocyte immunoglobulin-like receptor subfamily B member 4 (LILRB4), also known as ILT3, CD85k, and LIR-5, is a member of the leukocyte immunoglobulin-like receptor (LIR) family.
- LILRB4 functions as an inhibitory receptor that plays a critical role in down-regulating immune responses and establishing immune tolerance (PubMed:11875462). Related references: PMID:11875462
- It acts as a receptor for fibronectin (FN1; PubMed:34089617), apolipoprotein E (APOE; PubMed:30333625), and ALCAM/CD166 (PubMed:29263213). Related references: PMID:34089617, PMID:30333625, PMID:29263213
- Upon ligand binding, LILRB4 inhibits phosphorylation of cellular proteins and mobilization of intracellular calcium (PubMed:9151699). It specifically suppresses FCGR1A/CD64-mediated monocyte activation by inducing phosphatase-mediated down-regulation of kinases such as LCK, SYK, LAT, and ERK, thereby reducing TNF production (PubMed:19833736, PubMed:34089617). Related references: PMID:9151699, PMID:19833736, PMID:34089617
- LILRB4 modulates T cell responses by inhibiting T cell proliferation, inducing anergy, suppressing differentiation of IFNG-producing CD8+ cytotoxic T cells, and enhancing the generation of CD8+ T suppressor cells (PubMed:16493035, PubMed:19833736, PubMed:29263213). Related references: PMID:16493035, PMID:19833736, PMID:29263213
- The protein is localized on the cell membrane.
- It is expressed on monocytes, macrophages, dendritic cells, natural killer cells, and B cells (at protein level), with additional detection in the lung.
- LILRB4 contains immunoglobulin-like domains and disulfide bonds and undergoes phosphorylation, consistent with its role as an inhibitory receptor.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 22-259 of human LILRB4, which comprises the extracellular domain. Therefore, the antibody is expected to recognize the native receptor on the cell surface, making it suitable for flow cytometry applications.
- For flow cytometry, it is recommended to titrate the antibody to determine the optimal concentration for your specific cell type. Use an appropriate isotype control to assess non-specific binding.
- Consider validating the antibody in the relevant sample system, such as human peripheral blood mononuclear cells (PBMCs) or cell lines expressing LILRB4, under your experimental conditions.
- Ensure proper handling and storage of the antibody as per the manufacturer's instructions to maintain activity.
