Biological Background: LAMP2 Function and Localization
- LAMP2 is a heavily glycosylated lysosomal membrane glycoprotein containing a disulfide bond and is a member of the LAMP family.
- Acts as a direct inhibitor of the lysosomal proton channel TMEM175, thereby regulating lysosomal lumen pH and enabling optimal hydrolase activity. Related references: PMID:37390818
- Serves as a critical receptor for chaperone-mediated autophagy (CMA), binding cytosolic chaperone-recognized substrates (e.g., GAPDH, GPX4, NLRP3) and targeting them for lysosomal degradation. Related references: PMID:11082038 PMID:18644871 PMID:24880125 PMID:36586411
- Required for autophagosome-lysosome fusion; cells lacking LAMP2 fail to recruit STX17 to autophagosomes, preventing degradation of autophagic cargo. Related references: PMID:27628032
- Facilitates efficient MHC class II presentation of exogenous antigens by enabling lysosomal protein degradation, though it is not required for endogenous antigen presentation. Related references: PMID:15894275 PMID:20518820
- Supports FURIN-mediated cleavage of mumps virus fusion protein F by interacting with both FURIN and the unprocessed viral protein.
- Undergoes alternative splicing to produce three isoforms (LAMP-2A, -2B, -2C) with distinct tissue distributions; LAMP-2A is abundant in placenta/lung/liver, while LAMP-2B is broadly expressed, and LAMP-2C is enriched in the digestive tract and brain. Related references: PMID:26856698 PMID:7488019
- Localizes primarily to lysosome, endosome, and autophagosome membranes, and can also be detected at the cell surface.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the luminal domain (amino acids 29-375) of human LAMP2; the antibody likely recognizes an extracellular/luminal epitope accessible after permeabilization.
- For intracellular flow cytometry, ensure appropriate fixation and permeabilization optimized for the conjugate to maintain fluorescence signal.
- LAMP2 is expressed in many cell types; consider including appropriate positive (e.g., HeLa, Jurkat) and negative controls to confirm specificity in your experimental system.
- If cross-species reactivity is required, validate the antibody in the relevant non-human samples, as cross-reactivity may depend on epitope conservation.
- When combining LAMP2 detection with other lysosomal markers, verify compatibility of fixation and permeabilization protocols to preserve antigenicity and dye integrity.
