Biological Background: LAMP2 Function and Localization
- LAMP2 is a heavily glycosylated lysosomal membrane protein belonging to the LAMP family, characterized by a large luminal domain, a single transmembrane helix, and a short cytoplasmic tail.
- Functions as a critical regulator of lysosomal pH by directly inhibiting the proton channel TMEM175, thereby facilitating lysosomal acidification for optimal hydrolase activity. Related references: PMID:37390818
- Plays an essential role in chaperone-mediated autophagy (CMA), binding cytosolic proteins such as GAPDH, GPX4, NLRP3, and MLLT11, and targeting them for lysosomal degradation downstream of HSC70 recognition. Related references: PMID:11082038 PMID:18644871 PMID:24880125 PMID:36586411
- Required for macroautophagy by mediating the fusion of autophagosomes with lysosomes, likely through the recruitment of STX17 to autophagosomes. Related references: PMID:27628032
- Contributes to adaptive immunity by enabling efficient MHC class II-mediated presentation of exogenous antigens via its role in lysosomal protein degradation. Related references: PMID:15894275 PMID:20518820
- Alternative splicing produces three main isoforms (LAMP-2A, LAMP-2B, and LAMP-2C) with distinct tissue distribution patterns; LAMP-2A is highly expressed in placenta, lung, and liver, while LAMP-2B and LAMP-2C are more broadly expressed across tissues. Related references: PMID:26856698 PMID:7488019
- Localizes predominantly to the lysosome membrane, and is also found on endosome, cell, and autophagosome membranes.
- Associated with keywords such as alternative splicing, autophagy, glycogen storage disease, glycoprotein, disulfide bond, and transmembrane helix, reflecting its structural and functional diversity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 29–375 of human LAMP2, encompassing the entire luminal domain; thus, the antibody likely recognizes a conserved epitope present across all splice isoforms.
- Designed for intracellular flow cytometry; ensure proper cell permeabilization to allow access to the lysosomal membrane protein.
- For surface staining, note that LAMP2 can shuttle to the plasma membrane, but permeabilization will detect total LAMP2. Validate in your cell system to distinguish surface versus intracellular populations.
- As with all flow cytometry applications, titrate the antibody to determine the optimal concentration for your specific sample type and staining conditions.
