Biological Background: Human Kappa Light Chain Function and Localization
- The immunoglobulin kappa constant region (IGKC) is the constant part of the light chain in antibodies. It pairs with a variable domain to form the complete light chain and associates with a heavy chain to constitute an immunoglobulin monomer.
- Immunoglobulins are membrane-bound or secreted glycoproteins produced by B lymphocytes. Membrane-bound forms act as receptors that, upon antigen binding, trigger B cell clonal expansion and differentiation into antibody-secreting plasma cells. - The antigen binding site is formed by the variable domains of one heavy and one light chain. These variable domains are generated through V-(D)-J recombination and can undergo somatic hypermutation, allowing affinity maturation upon antigen exposure. - The kappa constant region is present in both secreted antibodies and as part of the membrane-bound B cell receptor (BCR). Its subcellular locations include secreted extracellular space and the plasma membrane.
- Structurally, the constant region contains conserved cysteine residues forming an intrachain disulfide bond, typical of the immunoglobulin domain fold. The theoretical molecular weight is ~11.8 kDa (UniProt), consistent with the observed 12 kDa in SDS-PAGE.
- IGKC is highly conserved among human allotypes (Km allotypes) and is involved in adaptive immunity. Mutations or imbalances in light chain production can lead to immunoglobulin deposition diseases.
Experimental Guidance and Technical Tips
- The immunogen covers the full-length human kappa constant region (amino acids 1-107), enabling broad recognition across all kappa light chain variants and allotypes.
- For Western blot (WB) under reducing conditions, a band at ~12 kDa (free light chain) or ~25 kDa (heavy + light chain) may be observed. Validate the specific band pattern in your sample type.
- In indirect immunofluorescence (IF-P), consider using appropriate fixation and embedding protocols. The antibody detects the constant region, so both surface and intracellular kappa light chain can be visualized.
- For flow cytometry (FC), ensure proper isotype controls and titrate the antibody for optimal staining of human B lineage cells or samples containing human immunoglobulins.
- For ELISA, the rabbit monoclonal format provides consistent batch-to-batch performance; the antibody can serve as a detection or capture component for human antibody quantification assays.
