Biological Background: IL-6R/CD126 Function and Localization
- Interleukin-6 receptor subunit alpha (IL-6R/CD126/gp80) is a transmembrane glycoprotein belonging to the immunoglobulin superfamily; a soluble form (sIL6R) is generated by alternative splicing.
- Part of the receptor for interleukin 6: binds IL6 with low affinity and requires association with IL6ST/gp130 for signal transduction, regulating immune responses, acute-phase reactions, and hematopoiesis (PubMed:28265003, PubMed:30995492, PubMed:31235509). Related references: PMID:28265003, PMID:30995492, PMID:31235509
- Membrane-bound IL6R mediates 'classic signaling', which is predominantly regenerative and anti-inflammatory; it also drives Th17 differentiation via 'cluster signaling' between dendritic cells and T cells.
- Soluble IL6R acts as an agonist: the IL6:sIL6R complex (hyper-IL6) enables 'trans-signaling' on cells lacking membrane IL6R, promotes VEGF production, and is causative for pro-inflammatory properties in chronic inflammatory diseases (PubMed:21990364, PubMed:12794819). Related references: PMID:21990364, PMID:12794819
- sIL6R also plays protective roles, being required for liver tissue regeneration after injury and regulating energy and glucose homeostasis in the central nervous system.
- Subcellular localization: the full-length receptor is a single-pass type I membrane protein; the soluble form is secreted.
- Expressed in peripheral blood mononuclear cells; sIL6R is detectable in urine and serum, with 1‑20% derived from alternative splicing (PubMed:28060820). Related references: PMID:28060820
- Post‑translational modifications include N‑linked glycosylation and disulfide bond formation within the extracellular immunoglobulin-like domain.
Experimental Guidance and Technical Tips
- The immunogen (aa 20‑358) covers the entire extracellular domain; the antibody may recognize both membrane‑bound IL‑6R and soluble sIL6R. Validate in the relevant sample system.
- This conjugate enables direct detection in flow cytometry without a secondary antibody. Confirm instrument compatibility (excitation ~650 nm, emission ~665 nm) and set compensation controls.
- For intracellular staining, optimize fixation and permeabilization conditions for your cell type to maintain epitope accessibility and minimize background.
- Use appropriate negative controls (e.g., isotype control, knockout/knockdown cells) to confirm specificity in your assay.
