Biological Background: Interleukin-5 Function and Localization
- Interleukin-5 (IL-5) is a homodimeric cytokine, also known as eosinophil differentiation factor (EDF) and T-cell replacing factor (TRF), encoded by the IL5 gene (GeneID: 3567).
- Plays a critical role in the survival, differentiation, and chemotaxis of eosinophils. Related references: PMID:2653458 / PMID:9010276
- Promotes growth, differentiation, and immunoglobulin production in activated and resting B-cells.
- Binds to a heterodimeric receptor composed of IL5RA and CSF2RB subunits. Related references: PMID:1495999 / PMID:22528658
- Receptor engagement activates kinases including LYN, SYK, and JAK2, propagating signals through RAS-MAPK and JAK-STAT5 pathways. Related references: PMID:7613138
- Secreted glycoprotein with intramolecular disulfide bonds; present in peripheral blood mononuclear cells.
- Post-translational modifications include N-linked glycosylation and disulfide-bond formation essential for bioactivity.
- Alternative splicing generates multiple isoforms; the canonical form is a 134-amino acid precursor with a 19-amino acid signal peptide, yielding a mature protein of ~15 kDa.
Experimental Guidance and Technical Tips
- The immunogen (aa 20–134) corresponds to the mature IL-5 protein; for Western blot, a band around 15 kDa under reducing conditions is expected; a ~30 kDa band may represent the covalent homodimer under non-reducing conditions.
- For immunofluorescence (IF/ICC), permeabilization may be required to detect intracellular or secreted IL-5; optimize fixation conditions.
- In ELISA, pilot experiments should determine optimal capture and detection conditions using recombinant IL-5 standards.
- Due to predicted wide cross-reactivity, this antibody may recognize IL-5 from multiple species; validate in the target species of interest.
- Store at recommended temperature and avoid repeated freeze-thaw cycles to maintain activity.
