- For Western blotting under reducing conditions, the IgM heavy chain migrates at approximately 75 kDa due to glycosylation, although the predicted unmodified weight is 49 kDa. Validate the band shift in your lysate.
- In immunofluorescence/ICC, surface IgM can be detected on live B cells; permeabilization may reveal intracellular pools. Optimize fixation and permeabilization for your cell type.
- Flow cytometry: this antibody is suitable for staining B cell surface IgM. Titrate the antibody and include appropriate isotype controls.
- ELISA: use this antibody as a detection reagent for human IgM in sandwich immunoassays. Cross‑reactivity with other species should be checked if extending to non‑human samples.
