Biological Background: IgD Function and Localization
- Immunoglobulin heavy constant delta (IgD) is the constant region of immunoglobulin delta heavy chains, existing as membrane-bound (mIgD) or secreted (sIgD) glycoproteins. Each IgD molecule possesses two antigen-binding sites formed by the variable domains of one heavy and one light chain.
- IgD serves as the major antigen receptor isotype on the surface of most peripheral B cells, coexpressed with IgM. Antigen binding to mIgD triggers B-cell activation, clonal expansion, and differentiation, and induces CD79A/CD79B phosphorylation by Src kinases. Related references: PMID:20176268, PMID:22158414
- The variable regions of IgD are generated by V(D)J recombination and can undergo somatic hypermutation, allowing affinity maturation upon antigen encounter. Related references: PMID:17576170, PMID:20176268
- Soluble IgD (sIgD) is found in serum at concentrations lower than IgG, IgA, and IgM but much higher than IgE. B-cell activation downregulates sIgD secretion.
- IgD is a potent inducer of pro-inflammatory cytokines such as TNF, IL1B, IL6, and IL10 from peripheral blood mononuclear cells, predominantly mediated by monocytes. Related references: PMID:10702483, PMID:11282392, PMID:8774350
- IgD is a glycoprotein with intra- and inter-chain disulfide bonds. Alternative splicing generates isoforms, and the protein weight of the constant region is approximately 42–47.5 kDa.
Experimental Guidance and Technical Tips
- For Western blot (WB): The antibody recognizes a band at approximately 42–47.5 kDa under reducing conditions. Verify expression in B-cell lines (e.g., Ramos or Daudi) or peripheral blood mononuclear cells.
- For flow cytometry (FC): Perform surface staining on viable human B cells; fixation and permeabilization may be required for intracellular detection of sIgD in activated cells.
- For ELISA: Use as a capture or detection antibody in sandwich ELISA formats for human IgD quantification; ensure specificity against other immunoglobulin isotypes by cross-testing.
- The immunogen covers the constant region (aa 99-384), suggesting the antibody detects all IgD isoforms sharing this sequence. Consider validation in the relevant sample system and application, as conditions may vary.
