Biological Background: IFN-alpha/beta R2 Function and Localization
- Interferon alpha/beta receptor 2 (IFNAR2), also known as IFNABR, IFNARB, or IFN-alpha-REC, is the ligand-binding subunit of the type I interferon receptor complex.
- Together with IFNAR1, it forms the heterodimeric receptor that mediates cellular responses to type I IFNs (alpha, beta, epsilon, omega, kappa) by activating the JAK-STAT signaling pathway. - Alternative splicing generates multiple isoforms, including a secreted form (isoform 3) found in urine that may modulate receptor activity. Related references: PMID:7759950, PMID:8181059
- IFNAR2 is predominantly localized to the cell membrane but can also be secreted. It is expressed in blood cells and detected in lymphoblastoid and fibrosarcoma cell lines.
- Post-translational modifications such as disulfide bond formation, glycosylation, and phosphorylation are important for IFNAR2 stability and function.
Experimental Guidance and Technical Tips
- This -conjugated monoclonal antibody is designed for flow cytometry detection of cell surface IFNAR2. The immunogen corresponds to the extracellular region (amino acids 28–237), ensuring specificity for the ligand-binding domain.
- For flow cytometry, titrate the antibody to determine the optimal concentration for your cell type. Use appropriate isotype controls and viability dyes to ensure accurate gating.
- Consider using IFN-alpha/beta-responsive human cell lines (e.g., lymphoblastoid or fibrosarcoma lines) as positive controls, and validate expression under stimulated vs. unstimulated conditions.
- The antibody cross-reacts with human IFNAR2; for non-human samples, verify cross-reactivity independently.
